Therapeutic Peptides and Proteins: Formulation, Processing, and Delivery Systems

As is the case with other pharmaceuticals, formulation development is one of the critical steps in developing a protein as a therapeutic product. Development of stable protein formulations may require even more resources and effort than conventional small molecule pharmaceuticals. Proteins typically have more stability issues as a result of their complexity and delicate structural stability. Fortunately, a great deal of research regarding protein stability has been conducted and this information is readily available in the literature reviewed by Manning et al. Ultimately, it would be ideal to be able to develop a pure pharmaceutical containing only the native protein. However, it is not practical to have only the native form of a protein in the formulation because the protein must be purified from a complex biological mixture containing a pool of other proteins which includes misfolded, denatured, and degraded forms of the same protein. Furthermore, a major challenge is to maintain the integrity of the purified protein during routine pharmaceutical processing, storage, handling, and delivery to the patient.

Pharmaceutical formulation , in pharmaceutics , is the process in which different chemical substances, including the active drug , are combined to produce a final medicinal product. The word formulation is often used in a way that includes dosage form. Formulation studies involve developing a preparation of the drug which is both stable and acceptable to the patient.

Approaches for improvement in therapeutic efficacy Improvements to formulations that either increase residence time or promote diffusion from the middle coat may be effective in overcoming the barriers to periocular delivery; nano-size formulations may provide superior diffusion; charge of formulations determines the interaction or diffusion process.

Further, these amino acids need not be adjacent to each other in the primary sequence. Each of the above issues significantly restricts the practical use of hydrogel-based drug delivery therapies in the clinic?

Reasonable efforts have been made to publish reliable data and information, and sources are indicated. Importantly, but the author and the publisher cannot assume formulatioj for the validity of all materials or for the consequence of their use. The appearance of a maximal pH was attributed to competing effects of pH on ascorbic acid. Reprinted material is quoted with permission, the conformational dynamics of the peptide in the glassy polymer exhibited a higher energy barrier between states than seen for the peptide in water. AK Banga. Taylor and Francis. The 10 chapters of Therapeutic Peptides and Proteins are well written and connect readers to the science-based observations and latest information available from different sources. One major accomplishment of this author is the citation of the original articles and the significance of contributions of the earliest investigators in the field of therapeutic peptides and proteins. The cross referencing is intense and topics within each chapter are well focused, with all relevant scientific information. I commend the author for his valiant efforts to single handedly collect and compose a vast information network.
